4. Results of a simulation of bacterial growth Fig. 5 shows the growth in pure culture of a theoretical organism similar to Escherichia coli. The simulator included provision for determining the moment of the end of the lag phase and it ended after 1 h and 49 min incubation. Fig. 7 (a and b) shows the total viable count and individual counts of the two strains. A substance slowly bacteriocidal for the sensitive strain was added at 12 h. After the end of the lag phase (3 h 20 min) both strains grew for a total of 8 h but the M-concentration (let us suppose a result of de-oxygenation of the medium) then began to exert an influence on both populations, the effect being most marked on the slower growing organism. After addition of the anti-bacterial substance the population of sensitive organisms declined steadily and about 2 h later, when numbers of the sensitive strain had fallen sufficiently, growth of the resistant strain recommenced reaching a maximum population after 32 h 30 min incubation.

Staining reactions include Gram stain, acid fastness, bipolar staining and the presence of metachromatic granules, and these can sometimes be divided into several grades. With all these characters the growth conditions and time of incubation should be standardized. If there are large changes during growth it may be permissible to score the results at two incubation times separately, although the sets of characters will not be entirely independent. It is best to base the scoring on averages of a number of organisms, and occasionally the degree of variation may also be usefully scored.
